Penn Immunologists Find a Molecule That Puts the ... - Health.am

We couldn?t live without our immune systems, always tuned to detect and eradicate invading pathogens and particles. But sometimes the immune response goes overboard, triggering autoimmune diseases like lupus, asthma or inflammatory bowel disease.

A new study led by University of Pennsylvania researchers has now identified a crucial signaling molecule involved in counterbalancing the immune system attack.

?The immune response is like driving a car,? said Christopher Hunter, professor and chair in the Department of Pathobiology in Penn?s School of Veterinary Medicine. ?You hit the accelerator and develop this response that?s required to protect you from a pathogen, but, unless you have a brake to guide the response, then you?ll just careen off the road and die because you can?t control the speed of the response.?

The research to characterize this immune system ?brake? was led by Hunter and Aisling O?Hara Hall, a doctoral candidate in the Immunology Graduate Group. Additional Penn collaborators included scientists from the Penn Genome Frontiers Institute?s Department of Biology and the Perelman School of Medicine?s Department of Medicine. Researchers from Merck Research Laboratories, the National Institute of Allergy and Infectious Disease, Harvard Medical School and Janssen Research and Development also contributed to the work, which was published in the journal Immunity.

"Healthy people have these cells - you have them, I have them - that are called Tregs,? or regulatory T cells, Hunter said. ?If you don?t have them you develop spontaneous inflammation and disease.?

Different forms of regulatory T cells operate as the brakes on various kinds of inflammation, but, until now, scientists hadn?t been certain of how these Tregs became specialized to do their particular jobs.

Hall, Hunter and colleagues decided to follow up on a molecule called IL-27. Scientists used to think IL-27 played a role in causing inflammation, but, in 2005, a team of Penn researchers, including Hunter, found the opposite; it was actually involved in suppressing inflammation. Thus, when mice that lack IL-27 are challenged with the parasite Toxoplasma gondii, they develop overwhelming inflammation.

?We never worked out how it did that, but it was a paradigm change at the time,? Hunter said.

In the new study, the researchers delved deeper into IL-27?s role. They found that exposing regulatory T cells to IL-27 promoted their ability to suppress a particular type of inflammation. The Penn-led team also demonstrated that they could rescue infected IL-27-deficient mice by giving them a transfusion of regulatory T cells. This finding suggests that IL-27 is required to produce the Treg cells that normally keep inflammatory responses in check during infection.

?Very surprisingly, we were able to show that the Tregs could ameliorate the pathology in this system,? Hall said. ?We don?t think this is the only mechanism by which IL-27 limits immune pathology, but it sheds light on one mechanism by which it could be functioning.?

Further experiments showed that Tregs express a different suite of genes in the presence of IL-27 as compared to another molecule that has been implicated in this process, interferon gamma, or IFN-?. The researchers? findings indicate that the two molecules have division of labor when it comes to suppressing inflammation: IL-27 seems to be important in helping control inflammation at the site of inflammation, whereas IFN-? appears more significant in the peripheral tissues.

?At the site of inflammation, where you?re getting your pathology, that?s where IL- 27 is important,? Hall said.

With a new understanding of how IL-27 may cause a class of Tregs to become specialized inflammation fighters, researchers have a new target for ameliorating the unwanted inflammation associated with all kinds of autoimmune conditions.

?Now we have a molecular signature that may be relevant in inflammatory bowel disease, in multiple sclerosis, in colitis and Crohn?s disease, in rheumatoid arthritis, in lupus,? Hunter said.

Next on tap, the team plans to study IL-27 in the context of asthma, lupus and arthritis.

Roos and Sara Cheery of the Penn Genome Frontiers Institute Department of Biology; Steven Reiner, formerly of the Penn Department of Medicine; Cristina M. Tato and Daniel Cua of Merck Research Laboratories; Yasmine Belkaid, Guillaume Oldenhove, Nicolas Bouladoux and John Grainger of the National Institute of Allergy and Infectious Disease; Laurence A. Turka of Harvard Medical School; and M. Merle Elloso of Janssen Research and Development.

WILLIAMSTOWN -- Twelve months ago, Adam Marske was toting a clipboard and watching Chris Cleary play quarterback for Williams College.

Cleary was sacked on Williams? second drive at Trinity, played in the third and then retired to the bench with an ankle injury. Marske played the rest of the game and came close to leading the Ephs back for an upset victory.

"It?s always a little bit weird to think back to that game," Marske said. "When we prepared [for today], we watched that game on film. It?s different to see how far we have come as a team. It brings back those feelings I had during that game, and how unexpected it was. I didn?t have any time to think."

Marske and the Ephs will host the Bantams this afternoon at Weston Field, one year after Trinity escaped with a 21-13 win. In that game a year ago, Marske brought the Ephs back to within a touchdown on a four-yard TD pass to Darren Hartwell with 54 seconds left. Williams linebacker Chris Cameron recovered the onside kick, but the Ephs were flagged for illegal procedure. That forced a rekick, which was recovered by the Bantams, who ran out the clock. In that game, Marske was 20 of 33 for 237 yards, two touchdowns and an interception.

The junior from LaGrange, Ill., has picked up where he left off. He was 22 of 27 for 277 yards, three touchdowns and one interception in last week?s 41-7 thrashing of Colby. Marske leads NESCAC in pass efficiency

and is 36 points ahead of second-place Jesse Warren of Wesleyan.

The Bantams come to town having beaten Williams seven out of the last 10 games. Trinity is 4-1 in the last five games, but the average margin of victory has been 5.8 points. The last blowout win in the series came in 2006 when the Ephs earned a 41-16 win. If you are keeping tabs, the only two wins by the Ephs in the last 10 years came in seasons where Williams ran the table and went 8-0.

The Ephs will start junior Marco Hernandez at tailback. Starter Alex Scyocurka bruised a shoulder last week, and while available, won?t start. Tom Wohlwender, who did not dress in the opener, will make his season debut against the Bantams. Greg Payton, who did not play against Colby, will be in uniform. Left tackle Cong Tien, who did not dress last week, will be available and it will be a game-time decision for the coaches if he plays.

Williams will be tested by junior running back Evan Bunker, who ran for 146 yards and two touchdowns in Trinity?s win over Bates last week. He is the younger brother of Eric Bunker, who ran for 132 yards and a touchdown in last year?s win over the Ephs. If Williams can control the line of scrimmage, quarterback Ryan Burgess will have to win the game for the Bantams. He was 13 of 26 for 161 yards, two touchdowns and two interceptions last week.

"They?re fast, they?re physical," said Kelton. "They?re very athletic. They have a good contingent of returners. They?re a typical Trinity team in that they?re strong at the running back position and have a good defense."

The team behind the addictive fruit-slashing game Fruit Ninja and its many variants have finally brought Jetpack Joyride to Google Play.

New virus not spreading easily between people: WHO

LONDON (Reuters) - A new and potentially fatal virus from the same family as SARS which was discovered in a patient in London last week appears not to spread easily from person to person, the World Health Organization (WHO) said on Friday.

In an update on the virus, which has so far killed a Saudi man and made a patient from Qatar critically ill, the United Nations health agency said it was working with international partners to understand the public health risk better.

"From the information available thus far, it appears that the novel coronavirus cannot be easily transmitted from person to person," it said in a statement.

The WHO put out a global alert on Sunday saying a new virus had infected a 49-year-old Qatari who had recently travelled to Saudi Arabia, where another man with the same virus had died.

The Qatari was described as critically ill on Tuesday and is being treated in a London hospital. No new confirmed cases of infection with the virus have since been reported, the WHO said.

The new virus shares some of the symptoms of SARS, or Severe Acute Respiratory Syndrome, another coronavirus, which emerged in China in 2002 and killed around a tenth of the 8,000 people it infected worldwide.

Both patients who have so far been confirmed with the new virus suffered kidney failure.

SEVERITY

"Given the severity of the two laboratory confirmed cases, WHO is continuing to monitor the situation in order to provide the appropriate response, expertise and support to its member states," the WHO statement said.

Scientists at the European Centre for Disease Prevention and Control (ECDC), which monitors disease in the European Union, said initial virology results and the separation in time of the only two confirmed cases suggest the infection may have developed from animals. Such diseases are known as zoonoses.

"(It) is quite probably of zoonotic origin and different in behaviour from SARS," the scientists wrote in a "rapid communication" study in the online journal Eurosurveillance.

Asked about transmission and the possibility of animal to human spread, WHO spokesman Glenn Thomas said investigations were continuing.

"But from the evidence we have, and given that there are only two cases confirmed so far and there was a distance and time distance between the two cases, (the) assumption is that it isn't easily transferable person to person," he told reporters.

The WHO's clinical guidance to its 194 member states says health workers should be alert to anyone with acute respiratory syndrome and requiring hospitalization who had been in the Middle East where the virus was found or in contact with a suspected or confirmed case within the previous 10 days.

The U.N. agency has not recommended any travel restrictions in connection with the new virus, but said it was working closely with Saudi authorities on health measures for Muslims making the haj pilgrimage to Mecca.

Health experts said rapid progress has already been made in figuring out the nature and genetic makeup of the new coronavirus, and in coming up with tests.

"We are developing with our partners sensitive and specific diagnostic assays and these should be available in the next few days," Thomas told a briefing at WHO headquarters in Geneva.

"If any national authority is concerned about a patient who is under investigation, if they want to contact us, we can put them in touch with these laboratories and provide initial tests for any cases which are suspicious," he added.

Sudan, South Sudan to resume oil exports, no wider deal

ADDIS ABABA (Reuters) - The leaders of Sudan and South Sudan reached a border security deal on Wednesday that will restart badly needed oil exports, but failed to solve the other main conflicts left over when Africa's largest country split last year.

The deal, reached after more than three weeks of negotiations, will throw both economies a lifeline. It should for now prevent any resumption of the kind of fighting that broke out along the border in April - the worst violence since South Sudan seceded in July 2011 under an agreement that ended decades of civil war.

But the neighbors still deeply distrust each other and have a history of failing to implement agreements.

Faced with the threat of U.N. sanctions and economic collapse, Sudanese President Omar Hassan al-Bashir and his South Sudanese counterpart Salva Kiir agreed after four days of summit talks to set up a demilitarized border buffer zone.

The deal will allow landlocked South Sudan to resume oil exports through Sudan, providing both battered economies with dollars. The south had shut down its entire 350,000 barrels a day of output in January after the countries argued about transit fees.

"The agreement will be signed tomorrow," El-Obeid Morawah, spokesman for Sudan's Foreign Ministry, told Reuters. Atif Keir, spokesman for the southern delegation, confirmed the deal.

But despite pressure from the African Union, the United States and other Western powers, the two sides failed to settle the fate of at least five disputed oil-producing regions along the border.

Tensions over the unmarked 1,800-km (1,200-mile) common border spilled over into fighting in April, when South Sudan's army briefly occupied the Heglig oilfield, vital to Sudan's economy.

"We will talk about the claimed areas in future talks," Morawah said.

NO DEAL ON ABYEI

The neighbors were also unable to reach a solution for the border region of Abyei, which has symbolic significance to both and is rich in grazing lands. Plans for a referendum have failed over the question of who should participate.

"The African Union will decide on a final solution," Keir said.

The security deal will be signed on Thursday, but it will take several months to restart southern oil production as the export pipelines were filled with water to prevent them becoming blocked, and some fields were damaged during fighting.

The U.N. Security Council had set a September 22 deadline for a broad deal to end hostilities but that was informally extended until the end of the Addis Ababa summit.

The African Union even postponed a signing ceremony for the border security deal at the last minute on Wednesday to make a last push for at least some agreement on Abyei, but to no avail.

The border deal provides for both armies to pull back 10 km (6 miles) from the frontier. Special arrangements will be made for a strategic strip of land called Mile-14, which is important to Arab tribes allied to Sudan.

But the lack of a permanent solution for the disputed regions will pose future risks to border security. Their fate may ultimately be settled only in international arbitration, a lengthy process.

Furthermore, AU mediator Thabo Mbeki failed to achieve tangible progress in indirect talks between Sudan and the rebel Sudan People's Liberation Movement-North (SPLM-North), which is fighting the Sudanese army in two areas bordering South Sudan.

The SPLM-North is part of an alliance with rebels from the western region of Darfur who want to topple Bashir. Sudan refuses direct talks as it accuses the south of supporting the SPLM-North, which controls some territory on the Sudanese side of the border.

Diplomats say each side supports rebels on the other side. South Sudan last weekend accused Sudan's army of having airdropped weapons to a militia fighting the government. Sudan denied this.

A deal reached last month to allow aid into rebel-controlled areas has not been implemented yet.

Canadian autoworkers OK Ford contract

TORONTO, Sept. 23 (UPI) -- Auto workers in Canada have approved a four-year contract with Ford Motor Co. by a wide margin, their union leaders said Sunday.

The Canadian Auto Workers said in a release posted on its website that the deal received a thumbs-up from 82 percent of the members who voted at ratification meetings Saturday and Sunday in Hamilton, Windsor and Brampton in Ontario.

"Our members at Ford recognize that in these uncertain economic times, some of the most important elements of a new collective agreement are future investment and improved job security," CAW President Ken Lewenza said. "This new agreement will ensure that our facilities are well-positioned for a strong future in the North American auto industry."

The CAW represents about 4,500 Ford workers. Voting took place at a series of ratification meetings Saturday and Sunday in Hamilton, Windsor and Brampton, Ontario.

The CAW has reached a tentative agreement with General Motors Co. and members are to vote on it Wednesday and Thursday.

The third Big Three automaker, Chrysler Group LLC, has yet to reach a deal with the union, though negotiations have continued beyond the expiration of the previous contract.

The CAW represents a total of about 16,500 workers at GM and Chrysler.

Pakistani bounty placed on anti-Islam filmmaker

PESHAWAR, Pakistan (Reuters) - A Pakistani minister offered $100,000 on Saturday to anyone who kills the maker of an online video which insults Islam, as sporadic protests rumbled on across parts of the Muslim world.

"I announce today that this blasphemer, this sinner who has spoken nonsense about the holy Prophet, anyone who murders him, I will reward him with $100,000," Railways Minister Ghulam Ahmad Bilour told a news conference, to applause.

"I invite the Taliban brothers and the al Qaeda brothers to join me in this blessed mission."

A spokesman for Pakistan's prime minister said the government disassociated itself from the minister's statement.

While many Muslim countries saw mostly peaceful protests on Friday, fifteen people were killed in Pakistan during demonstrations over the video.

People involved in the film, an amateurish 13-minute clip of which was posted on YouTube, have said it was made by a 55-year-old California man, Nakoula Basseley Nakoula.

Nakoula has not returned to his home in the Los Angeles suburb of Cerritos since leaving voluntarily to be interviewed by federal authorities. His family has since gone into hiding.

In the Bangladeshi capital Dhaka on Saturday, thousands of Islamist activists clashed with police who used batons and teargas to clear an unauthorized protest. In Kano, northern Nigeria's biggest city, Shi'ite Muslims burned American flags, but their protest passed off peacefully.

The demonstrations were less widespread than on Friday, but showed anger still simmered around the world against the film and other insults against Islam in the West, including cartoons published by a French satirical magazine.

Showing continued nervousness among Western governments, German Foreign Minister Guido Westerwelle called on Muslim countries to protect foreign embassies.

"The governments in host countries have the unconditional obligation to protect foreign missions. If that doesn't happen, we will emphatically criticize that and if it still doesn't happen it won't go without consequences," he told the Welt am Sonntag newspaper on Sunday

Germany's embassy in Sudan was stormed on September 14 as was the U.S. mission in the capital Khartoum where there were deadly clashes between police and protesters against the film.

In the Libyan city of Benghazi, a crowd forced out an Islamist militia some U.S. officials blame for a deadly attack on the U.S. consulate during one of the first protests, on September 11.

Ansar al-Sharia, which denies it was involved in the attack that killed U.S. Ambassador Christopher Stevens and three other Americans, quit the city after its base was stormed by Libyans angry at armed groups that control parts of the country.

That might go some way to vindicate U.S. President Barack Obama's faith in Libya's nascent democracy where Ambassador Christopher Stevens had worked to help rebels oust Muammar Gaddafi only to be killed in a surge of anti-Americanism.

"It's the view of this administration that it's a pretty clear sign from the Libyan people that they're not going to trade the tyranny of a dictator for the tyranny of the mob," said White House spokesman Josh Earnest.

"It's also an indication that the Libyan people are not comfortable with the voices of a few extremists and those who advocate and perpetrate violence, to drown out the voices and aspirations of the Libyan people." [ID:nL5E8KM49W]

In Egypt, the leader of Egypt's main ultra orthodox Islamist party, that shares power with the more moderate Muslim Brotherhood, said the film and the French cartoons were part of a rise of anti-Islamic actions since the Arab spring revolts.

"A new reality in the Middle East has emerged after the toppling of autocratic regime of Hosni Mubarak and others through democratic elections that brought newly-elected Islamist governments," Emad Abdel Ghaffour, leader of the Salafist Nour Party, told Reuters.

"There are interest groups who seek to escalate hatred to show newly-elected governments and their Muslim electorate as undemocratic," he said.

Nour, whose party is the second largest in parliament and plays a formidable force in Egypt's new politics, said President Mohamed Mursi should demand "legislation or a resolution to criminalize "contempt of Islam as a religion and its Prophet" at the U.N. General Assembly next week.

On?a day that shattered records ? and perhaps the confidence of electro-pop duo LMFAO ? Guinness World Records announced that the video for "Gangnam Style," by Korean rapper PSY is the most liked video in YouTube history.

Gaining 2,141,758 user "likes" since its July 15 YouTube debut, "Gangnam Style"?topples previous record holder, "Party Rock" by LMFAO, which released a statement on Friday saying the two were taking time off. (No mention on whether the hiatus is connected to the record loss ? probably not, but there you go.)

Meanwhile, everything keeps coming up PSY, with Friday's announcement for Guinness adding to the K-Pop star's growing list?wins, including?"Gangnam Style's" status as the?first K-Pop song to hit No. 1 on iTunes.

"Having been the 'Have you seen this?!' video of the last two months across the Web, it's great to be able to award a record for this tremendously popular video," Dan Barrett, Guinness World Records community manager, said in a statement. "In years past, it was unthinkable that something? would be viewed a hundred million times, and now 'Gangnam Style' has achieved more than twice this figure in just 3 months on YouTube."

Along with its more than 2.1 million "likes," "Gangnam Style," also has an obligatory 100K dislikes, and more than 227 million views ... and that's not including all the viral tributes.

My sons hear me talk about why these words are truly hurtful, soul-dulling words. At seven, Nate understands. When he hears others say one of them he gasps and points with such drama you would think a coup d??tat was afoot. He reacts the same way when someone has eaten the last brownie but painstakingly covered the empty pan with foil and left it on the counter. Ahem?

This week, I?ve decided I am adding a word to the lexicon of evil. FAIR. Fair is hitting my list.

There is no such thing as fair. Nothing is fair, nothing ever will be fair. Things are or they are not, but absolute, unchallenged, all-inclusive equity simply does not exist. And I am sick to death of trying to correct the practical ?not fairs? like ?it isn?t fair that this boy in my class has a summer house AND a winter cabin!? or ?it is only fair if both Sam and me get the exact same vitamin shape every single day.? Not to mention the absurd not fairs like ?it isn?t fair that Spiderman can shoot webs and I can?t? and ?hey no fair that you got up before me.? Really? To whom is that not fair?

My real issue with fair is not the annoying statements or my inane need to address each of them with comments about what you DO have and CAN do, or my core need to instill a radical level of gratitude in a 2- and 7-year-old whose brains may simply not be ready. My real issue is that I think that labeling things, people, actions, as fair or not is a way to separate oneself from others. It is a way to keep some folks out of the circle and some in. It is a way for kids especially to point out difference and for adults to perpetuate a sense of not having or not being something that they put outside of their own control. Basically it puts a word to self pity or self aggrandizing or the inability to create change. All of which I refuse to buy into. And more importantly, I refuse, flat out refuse, to let my sons buy into. Something you want? Is it really that important and if so, what are you willing and able to do to have it? Want to be something? Figure out a way to make it happen. See an inequity? What can you do to fix it? Don?t just slap a word on it and throw up your hands to the universe and pout.

When I respond with these comments and questions to statements like the one for multiple homes or for web-spinning skills, we create conversations about the real or lack of importance of these things. Most of the time our discussion ends with a sense that that thing or process is really not that important in the scheme of our lives as a family or Nate?s place in the world. I want him to find another way to articulate what he envies or fantasizes about without labeling it with the judgment-heavy ?fair.? That doesn?t get us anywhere. There is nowhere to go from that label except to the awfully lonely land of ?us and them.?

So I?m taking a stand. FAIR is a word that hurts and damages. It is going on the list. Express your desires and need for inclusion in other words because that one doesn?t help any of us be better. And we can be so much better.

How bad is it for Mitt Romney right now? Politico's lead story has advisers and friends and "a person who recently was alone with Romney" preemptively chucking dirt on his campaign. On a morning conference call, advisers made much of how Romney was only three points behind President Obama in the Gallup tracking poll. It was thin gruel.
